Abstract
ABSTRACT Purpose. Erectile dysfunction (ED) after radical prostatectomy (RP) for prostate cancer has been known to be caused by cavernous nerve (CN) injury. This study was designed to investigate the therapeutic effect of Ad-Smad 7 vector (ASV), an inhibitory Smad that blocks transforming growth factor-? signaling pathway, in an mouse model of cavernous nerve injury. Materials and Methods. Twelve-week-old C57BL/6J mice were divided into 4 groups (N = 6 per group): Sham operation, bilateral CN crush injury(CNI), bilateral CN crush injury with Ad-Lac vector (ALV), and bilateral CN crush injury with ASV group. Bilateral CN crush injury with ASV group was analyzed as a function of dose (1x107, 1x108, 2x108 and 1x109 virus particles [vp] in 20 ?l of phosphate-buffered saline). Erectile function was measured by electrical stimulation of the cavernous nerve at 2 weeks after CN crush injury. The penis was then harvested and stained with antibodies to platelet/endothelial cell adhesion
